Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
Regulatory approval in the United States came in 2010, when the Food and Drug Administration cleared the peptide for the reduction of excess abdominal fat in adults with HIV infection and associated lipodystrophy. The decision rested mainly on two randomized phase 3 trials that enrolled roughly eight hundred patients and ran for twenty-six weeks. Participants receiving active drug showed substantially greater declines in visceral adipose tissue than those receiving placebo, while total body weight changed comparatively little. A reformulated presentation was later approved, and the product has remained a niche therapy rather than a general weight-loss agent.

Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Quantitation of the peptide relies mainly on reversed-phase high-performance liquid chromatography with ultraviolet detection, typically at 214 nanometers, where the peptide bond absorbs. Identity is confirmed by mass spectrometry, most often electrospray ionization coupled to liquid chromatography, and by peptide mapping after enzymatic digestion. Because related impurities differ only slightly in sequence or modification, method development emphasizes resolution rather than speed. Purity is usually reported as a percentage of the main peak area, with individual impurities listed separately when they exceed a defined reporting threshold.
Stability testing examines how the molecule changes under controlled stress. Thermal stress, light exposure, and extremes of pH are applied separately so that each degradation route can be attributed to a specific cause. The main observed changes are oxidation, deamidation, and aggregation into dimers or higher-order species. Accelerated studies at elevated temperature are used to estimate behavior over longer periods, though such extrapolation carries uncertainty. For a lyophilized powder, residual moisture and the choice of bulking agent strongly influence how quickly these changes appear.
Practical handling centers on limiting moisture, oxygen, and temperature excursions. Lyophilized material is generally held at or below minus twenty degrees Celsius, protected from light and kept sealed until use. Once reconstituted, solutions are typically kept cold and used within a short window because hydrolysis and microbial growth both accelerate in liquid form. Repeated freeze-thaw cycles are avoided, since they promote aggregation. Vial contents should be inspected for particulates and clarity before analysis, and working aliquots are prepared to reduce the number of times the stock is opened.
Amylin analogues can both reduce energy intake and increase expenditure and can usefully be combined with leptin analogues for synergistic effect. The dual amylin and calcitonin receptor agonist cagrilintide, in combination with semaglutide, was more effective than semaglutide alone in promoting weight loss in clinical trials. Glucagon receptor agonists both reduce energy intake and increase energy expenditure in humans. They can cause hyperglycemia so it is recommended to combine them with a hypoglycemic drug, such as a GLP-1 or GIP receptor agonist.
Brenipatide (INNTooltip International Nonproprietary Name, USANTooltip United States Adopted Name; developmental code name LY-3537031) is a dual agonist of glucagon-like peptide-1 (GLP-1) receptors and gastric inhibitory polypeptide (GIP) receptors. Brenipatide is under development by Eli Lilly and Company for the treatment of alcoholism, bipolar disorder, asthma, smoking withdrawal, cardiovascular disorders, liver disorders, metabolic disorders, and obesity. It is taken by subcutaneous injection once per month. The drug has a longer elimination half-life than tirzepatide or retatrutide. As of December 2025, it was in phase 3 clinical trials for alcoholism and bipolar disorder, phase 2 trials for asthma and smoking withdrawal, and phase 1 trials for cardiovascular disorders, liver disorders, metabolic disorders, and obesity. Actin, gamma-enteric smooth muscle is a protein that in humans is encoded by the ACTG2 gene. Actins are highly conserved proteins that are involved in various types of cell motility, and maintenance of the cytoskeleton. In vertebrates, three main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins co-exist in most cell types as components of the cytoskeleton, and as mediators of internal cell motility. Actin, gamma 2, encoded by this gene, is a smooth muscle actin found in enteric tissues. ACTG2 has been shown to interact with Emerin. Human ACTG2 genome location and ACTG2 gene details page in the UCSC Genome Browser.

Reticulocytes are newly produced red blood cells. They are slightly larger than totally mature red blood cells, and have some residual ribosomal RNA. The presence of RNA allows a visible blue stain to bind or, in the case of fluorescent dye, result in a different brightness. This allows them to be detected and counted as a distinct population. The RPI is used to assess whether the bone marrow is producing an appropriate response to an anemic state. Reticulocyte production should increase in response to any loss of red blood cells. It should increase within 2–3 days of a major acute hemorrhage, for instance, and reach its peak in 6–10 days. If reticulocyte production is not raised in response to anemia, then the anemia may be due to an acute cause with insufficient time to compensate, or there is a defect with red blood cell production in the bone marrow. Marrow defects include nutritional deficiencies (i.e. iron, folate, or B12) or insufficient erythropoietin, the stimulus for red blood cell production. Reticulocytopenia, or "aplastic crisis", is the medical term for an abnormal decrease of reticulocytes in the body The reticulocyte production index may find new use as a more reliable detector of erythropoietin-doping in athletes. The use of this method is referred to as "biological passport."
Ethylhexyl palmitate, also known as octyl palmitate, is the fatty acid ester derived from 2-ethylhexanol and palmitic acid. It is frequently utilized in cosmetic formulations. Ethylhexyl palmitate is a clear, colorless liquid with a slightly fatty odor at room temperature. The ester is synthesized by reacting palmitic acid and 2-ethylhexanol in the presence of an acid catalyst. Ethylhexyl palmitate is used in cosmetic formulations as a solvent, carrying agent, pigment wetting agent, fragrance fixative and emollient. Its dry-slip skinfeel is similar to some silicone derivatives.
90. ArXiv [Preprint]. 2026 Jul 29:arXiv:2605.17186v2. Operator splitting for exploiting linear-rate closure in solving infinite ODE hierarchies. Chang JC. We introduce an operator-splitting method for infinite hierarchies of linear ordinary differential equations (ODEs) indexed by nonnegative integers. When the coupling coefficients depend linearly on the count index, an exact transformation closes the equations on finite count-index windows without an upper-boundary value. For more general hierarchies, Strang splitting applies the linear-rate closure during the linear-rate substeps and a conventional capped solver to the remainder. We derive the closure from generating functions and the method of characteristics and extend it to multi-indexed systems. The derivation requires neither positivity nor mass conservation, so it applies to a wider class of systems than the examplar stochastic models presented here. We discuss branching processes, stochastic predator-prey dynamics, the Schlögl chemical kinetics model, and a telegraph model for gene expression. Through numerical experiments and computational cost analyses we demonstrate that our operator splitting method is typically advantageous for solving large scale systems in terms of memory usage and computational time, while retaining accuracy competitive with finite state projection (FSP) methods. The overall fold of Acutolysin A is composed of a twisted β-sheet core flanked by α-helices, forming the characteristic metzincin architecture. Central to this fold is the conserved “Met-turn”, a methionine-containing structural motif that stabilizes the active-site configuration. The three disulfide bonds in AaH I (Cys117–Cys197, Cys159–Cys181, and Cys157–Cys164) are strategically positioned to maintain this fold under physiological conditions and to resist thermal or proteolytic degradation. These disulfide linkages play a crucial role in preserving the shape of the catalytic cleft, ensuring maximal enzymatic activity even in harsh extracellular environments. At the active site is the HELGHNLGLH metalloproteinase motif, which binds a catalytic zinc ion in a tetrahedral geometry. Three histidine residues coordinate the zinc atom, while the fourth ligand is either a water molecule or hydroxide ion, which acts as the nucleophile in peptide bond hydrolysis. The active-site cleft forms a deep groove that accommodates collagen and laminin fibers, aligning them precisely for cleavage. This structural arrangement explains the exceptional potency of AaH I in degrading basement membranes.

The predecessor of modern countercurrent chromatography theory and practice was countercurrent distribution (CCD). The theory of CCD was described in the 1930s by Randall and Longtin. Archer Martin and Richard Laurence Millington Synge developed the methodology further during the 1940s. Finally, Lyman C. Craig introduced the Craig countercurrent distribution apparatus in 1944 which made CCD practical for laboratory work. CCD was used to separate a wide variety of useful compounds for several decades.
The right side of a positive-sensed AAV genome encodes overlapping sequences of three capsid proteins, VP1, VP2 and VP3, and two accessory proteins, MAAP & AAP, which start from one promoter, designated p40. The molecular weights of these proteins are 87, 72 and 62 kiloDaltons, respectively. The AAV capsid is composed of a mixture of VP1, VP2, and VP3 totaling 60 monomers arranged in icosahedral symmetry in a ratio of 1:1:10, with an empty mass of approximately 3.8 MDa. The crystal structure of the VP3 protein was determined by Xie, Bue, et al.

It is a laboratory-made peptide of forty-four amino acids whose sequence matches human growth hormone-releasing hormone, with a modified amino terminus. The modification is a short unsaturated fatty acid chain attached to the first residue. This change slows enzymatic breakdown and lengthens the time the peptide stays active in circulation.
